    Antibiotics are well known to be use for systemic therapy via parenteral as well as topical administration for bacterial infections. However, owing to their short half-life, most antibiotics and the conventional drug delivery systems, limits the pharmacokinetic profile of the drugs. This may display toxicity and narrow therapeutic effect to the targeted areas. This study reports the preparation and physicochemical characterization of chitosan crosslinked dialdehyde xanthan gum gels semi-interpenetrated with hydroxypropyl methylcellulose for the controlled delivery of minocycline and rifampicin antibiotic drugs. The prepared gels were characterized by FTIR, SEM, TGA, mechanical and rheology methods. In addition, gelation kinetics, swelling, degradation and drug release were studied under simulated stomach and plasma physiological conditions of pH 2.0 and 7.4, respectively, at 37 °C. The results showed more than 50% cumulative release within 24 h for all investigated antibiotic drugs. Cytocompatibility by MIT assay using mouse embryonic fibroblast cell lines were tested and cell viability was determined to be ????80%, confirrning the gels are non-toxic. Finally, the antibacterial effect of loaded gels was evaluated against Gram-negative and positive bacteria (Escherichia coli, Staphylococcus aureus and Klebsiella pneumonia). The observed antibacterial effect correlated well with swelling and drug release results. Overall, our results demonstrated the produced gels as promising materials for controlled delivery drug molecules for biomedical applications.
