Bone defects resulting from trauma, infection, or cancer present significant challenges in orthopaedic medicine, especially when they reach critical sizes, preventing spontaneous healing. Traditional treatments, such as autografts or allografts, often come with limitations, including donor site morbidity and limited availability. This study introduces a novel approach using chitosan-based hydrogels integrated with polypyrrole (PPy) nanoparticles and bone morphogenetic protein-2 (BMP-2).A water-soluble half N-acetylated chitosan (SCN) was used for its solubility at physiological pH while preserving its bioactive properties. Incorporating PPy not only improves the mechanical properties of the hydrogel but also provides antibacterial and anti-inflammatory effects critical for bone healing. Additionally, microspheres loaded with BMP-2 were included to stimulate osteogenesis and enhance healing processes. As a crosslinker, 2,3-dialdehyde cellulose (DAC) was used to covalently anchor PPy with SCN to ensure stability and prevent leaching of nanoparticles.The physicochemical characterisation of prepared hydrogels was evaluated by analysing viscoelastic properties and SEM analysis. The hydrogels exhibit shear-thinning behaviour, allowing them to fill irregular bone defects effectively through minimally invasive procedures.Initial in vitro tests will be conducted to evaluate the biocompatibility of the developed hydrogels. Later, the in vivo experiments on mice will be performed to analyse anti-inflammatory effects in the early stage of wound healing and their influence on bone regeneration.The synergistic effect of PPy’s anti-inflammatory activity, SCN’s enhanced biocompatibility, and BMP-2’s osteoinductive properties are expected to promote efficient bone regeneration. This hydrogel system offers a promising solution for treating critical-size bone defects, potentially overcoming the limitations of traditional treatments.
