Machine Learning Assisted Disentanglement of Complexity of Phytochrome Excited States
Anotace anglicky
The project aims to reveal the ultrafast photoinduced processes and underlie the mechanism of photoactivation in bacteriophytochrome BPhy protein samples. BPhys are an optimal model for studying the phytochrome protein family due to their photostability and reverse light-switching capabilities. These proteins play a crucial role in detecting and responding to environmental light, showing great potential in the development of new light-switching tools that can be used to control molecular functions. The problem will be jointly faced experimentally at the University of South Bohemia and computationally at the Chimie ParisTech. The primary objectives can be divided into two parts. The experimental part will utilize transient absorption spectroscopy to investigate the ultrafast dynamics and intermediate states during the photoconversion of BPhys. This involves studying various protein constructs and mutants to understand the structural changes and excited-state photophysics, such as photoisomerization and charge transfer reactions, following photon absorption. The computational project part will employ machine learning techniques to interpret experimental data, predict spectra for different experimental conditions, and identify key correlations between molecular structures and photophysical properties. This will enhance the understanding of the protein's response to light and will help in designing future experiments. Once the data are collected both experimentally and theoretically, these will be combined to train machine learning models that will be able to optimize laboratory performances, find correlations among data, and predict experimental signals. Through these integrated experimental and computational approaches, the project aims to provide comprehensive insights into the mechanisms of light-induced signal transduction in phytochromes, facilitating their potential applications in optogenetics and fluorescent probes.
