Anther and microspore cultures in flax, their development and evaluation of their efficiencies

Anotace anglicky

The project is focused on improvement of anther culture method and on development of microspore culture method with the aim to include more efficient one into flax improvement. Some factors affecting callogenesis in anthers and organogenesis in anther calli will be studied to find their the most suitable values for regeneration of haploid or dihaploid plants. Following factors will be studied in the anther culture: auxins in callus inducing medium, cytokinins and their changes in following subcultures,changes in culture medium nitrogen levels in following subcultures, light growth conditions of donor plants (light intensity and day length), some mitotic active compounds (colchicine, oryzalin), several different carbohydrates as a source of carbon andenergy. Variability in some morphological features in progenies of anther culture derived plants, initial material and material after classical selection as well will be compared.
