Nano-Oscillator Mass Photometry: Mass and Charge as Keys for Deciphering Biomolecular Pathways

"Proteins are performing essential functions in cells, thereby enabling life. Our 20,000 genes encode proteins that undergo posttranslational modifications (PTMs), potentially generating up to 100 modified forms. PTMs are linked to cardiovascular diseases, cancer, and neurodegenerative disorders such as Alzheimer’s or Parkinson’s. Understanding PTMs and their impact on protein-protein interactions is thus vital for advancing new therapeutics. Studying molecular pathways associated with PTMs is challenging because the majority of current methods do not operate in solution and struggle with PTM-associated heterogeneity. Our goal is to transform our ability to study proteins and their modifications by developing a platform technology capable of measuring not only the mass of single biomolecules in solution, but also their charge. This will enable us to differentiate proteoforms even for tiny mass differences, because PTMs such as phosphorylation and acetylation or sialylation come with significant modifications to overall molecular charge. To achieve this, we will create electrically driven nano-oscillators integrated with mass photometry. We will leverage the physics of a charged mass on a spring in an electric field, monitored by light scattering and changes to its oscillations upon biomolecular interactions. Specifically, we will: 1. Develop universal surface passivation that enables long-duration measurements under high-power illumination 2. Engineer specific biomolecule capture mechanisms. 3. Design low-scattering microelectrodes. 4. Create nanoparticles with a defined scattering response that do not obscure the protein signal. 5. Exploit real-time mass and charge measurements to investigate the molecular pathways of PTMs of cancer-related proteins, such as soluble p53 and membrane-bound EGFR, ultimately redefining the current understanding of PTMs related to viral entry glycoproteins and phase separation proteins."
