    Trypanosomatid biochemistry: an insight from the neglected groups

    Family Trypanosomatidae is a species-rich group of protists parasitizing a wide range of hosts. Several genera are pathogenic to vertebrates, animals, and plants and, thus, they are attractive for research and we have acquired a substantial amount of information about them. The infamously known Trypanosoma brucei causes the African sleeping sickness in human and nagana in livestock, while Leishmania mexicana is the causal agent of cutaneous leishmaniasis. Pathogenicity of trypanosomatids is associated with their shuttling between two hosts (dixenous species). However, most of the trypanosomatids are restricted to the invertebrate host (monoxenous trypanosomatids). They are harmless to human and, therefore, were effectively neglected for many years. Dixenous lifestyle evolved from monoxenous and requires many adaptations due to the environmental changes when parasite faces the temperature and nutrient alteration. Studying monoxenous species capable to reproduce at a wide temperature range may therefore help to better understand the evolution of disease-causing species. But there are certainly other properties of monoxenous trypanosomatids, which are associated with the transition to a dixenous lifestyle. For species studied to this date a high variability in mitochondrial enzymatic activities even among the closely related flagellates has been reported. It was especially pronounced for enzymes of oxidative phosphorylation. A common feature of all studied species is that the low or missing activity of one pathway is compensated by an increase of the other one. A better understanding of the energetic metabolism and phylogeny of more trypanosomatids is, therefore, essential to understand they transition to the dixenous lifestyle associated with pathogenicity. Further research will provide additional data potentially useful in applied medical research. Improving the knowledge of the metabolism of these parasites can, therefore, help to combat the damages they cause.

    V rámci projektu LUASK22033 „Biochemie trypanosomatid: pohled z opomíjených skupin“ jsme sekvenovali genomy a transkriptomy mnoha dosud nepopsaných druhů. To nám umožnilo provést in silico analýzu jejich metabolismu a potvrdit některé předpovědi v laboratoři. Výsledkem tohoto společného projektu bylo 9 vědeckých publikací, včetně několika článků ve vysoce uznávaných prestižních časopisech.
