Orthopaedic surgeries like total hip and knee arthroplasty play a crucial role in restoring joint function for individuals with osteoarthritis. Deep bacterial infection is one of the most serious complications of orthopedic implants. This complication results in pain, swelling, redness, and fistulation in the implant area. An infectious complication of an orthopedic implant requires long-term and demanding treatment, often with repeated surgical procedures, and can lead to serious consequences such as implant failure, sepsis, and even death. The early detection of complications is of key importance for efficient therapy. The objective of this project is to design, develop, and test a new generation of conceptually new multi-criterial sensors that can monitor inflammation and detect infections early around orthopaedic implants. These sensors will enable simultaneous potentiometric sensing of inflammation- and infection-related analytes (such as pH, reactive oxygen species, Ca2+, bacterial endotoxin, or defensin) at physiologically relevant concentrations in body fluids. This simultaneous detection of multiple analytes will provide a comprehensive understanding of pathophysiological processes on-site. The sensor, based on novel materials, will be used for constructing devices to detect inflammation-related analytes in body fluids, such as synovial fluid, during surgery directly in the operating room. Alternatively, the sensor´s electrodes could be miniaturized and integrated into an implant with a wireless data transfer device, allowing non-invasive, in situ sensing without the need for additional intervention. This project stands out as it combines clinical experience, research, and technical development.
