    Genetic characterization of important regional Norway spruce populations in the Czech Republic

    Presented work summarizes the basic genetic characteristics of 12 Norway spruce populations in the Czech Republic obtained by DNA analysis. For polymorphism verification of selected Norway spruce populations, method of using nuclear microsatellites was chosen. In order to both determine the genetic characteristics of selected populations and verify the level of their genetic diversity, mutual genetic distances and heterozygosity through 9 nuclear microsatellite markers were used. The fragmentation analysis by the genetic analyser were performed on the amplification products of analysed loci. The data for assessing the genetic characteristics of the Norway spruce populations were processed using the GenAlEx 6.5 statistical program. The evaluation of genetic distances between populations studied was calculated based on Nei's standard genetic distance, the values were processed by Principal Coordinate Analysis (PCoA) and Cluster Analysis (CLU). The results were clearly shown using the map. All analysed populations reported higher values of genetic diversity. Differentiation between populations is not inconsiderable and indicates a structuring of Norway spruce populations. Observed heterozygotes ranged 65–82%. Significant differences between populations were reflected in the frequency of unique alleles. The genetic distances of the populations approximately correspond to their geographic position. This results could be refined by applying new methods of DNA analyses, or taking into account more populations, SSR markers, and trees, in future.
