Photosynthetic characteristics and molecular basis of resistance to atrazine in a Czech biotype of redroot pigweed

A suspected atrazine-resistant biotype of Amaranthus retroflexus L. was found at the railway station in Prague-Bubny. Greenhouse experiments were performed to confirm resistance to atrazine in this weed biotype. The mechanism of resistance was determinedon the basis of chlorophyll fluorescence, Hill reaction activity and sequencing of the psbA gene. Results from chlorophyll fluorescence and photochemical activity assays confirmed that the Amaranthus retroflexus biotype from the Prague-Bubny railway station was resistant to atrazine and suggested that the resistance was site-of-action mediated. Sequence analysis was conducted for the psbA gene that codes for the target site of Dl protein. A mutation in the psbA gene was identified that coded for an amino acid substitution of glycine for serine at residue 264 of the Dl protein.
