Streptococcus suis is an economically significant pathogen that causes high morbidity and mortality negatively affecting the swine industry worldwide. Effective vaccines against S. suis have not been developed yet. Currently, only autogenous vaccines are available, but their protection is limited. In this study, we aim to develop experimental vaccines that would protect pigs against S. suis. Two types of conjugated vaccines were prepared by chemical conjugation of capsular polysaccharide (CPS), isolated from S. suis, to either ovalbumin (OVA) or cross-reactive material 197 (CRM197) as a carrier proteins. Immunogenicity of prepared conjugated vaccines was tested on mouse model. Six groups of ten mice were vaccinated with OVA and CRM197 proteins alone, with the CPS-OVA and CPS-CRM197 conjugates, with CPS alone and last group of ten mice were vaccinated with DPBS as a control group. The presence of antibodies in serum of immunized mice was confirmed by the western blot method, while ELISA method was then used to determine the concentration of antibodies. ELISA proved that groups of mice immunized with the conjugates induced a high immune response to the capsular polysaccharide as antigen, whereas the group of mice immunized with capsular polysaccharide alone did not induce any immune response. Groups of mice immunized with the conjugates also induced a greater immune response to the respective carrier proteins than groups of mice immunized with these proteins only. These experimental conjugate vaccines will be used for immunization and challenge of pigs. This project was supported by the Ministry of Agriculture (QK1810193 and RO0518).
