This chapter summarizes the current knowledge about the localization and function of substance P in the heart and blood vessels, as well as its significance in the central regulation of the functions of the cardiovascular system. Substance P is a prominent member of the tachykinin family, which acts to target tissues through neurokinin receptors. In the heart, substance P mainly acts on pacemaker cells and the intracardiac nervous system, through which it regulates cardiac functions. Furthermore, it affects the vasomotion of the coronary vessels as well as vessels of the systemic circulation. Involvement of substance P in the central regulation of cardiovascular functions includes the transmission of sensory information from vessels to cardiovascular centers in the brain stem and spinal cord, and also the regulation of their activity by the transmission of signals from other parts of the brain.
