Hydrophilic polymer conjugates bearing protoporphyrin IX precursors for photodynamic therapy of tumors

  • Popis výsledku v původním jazyce

Photodynamic therapy is an advantageous approach in anticancer treatment, mostly due to its non-invasive character. It consists of three main components - photosensitizer, light and oxygen. After photosensitizer administration, irradiation of the tumor tissue with light of appropriate wavelength takes place resulting in singlet oxygen production and tumor death. Photosensitizers are molecules based predominantly on porphyrins or similar structures with protoporphyrin IX (PPIX) being an endogenous photosensitizer. PPIX is usually administered in form of a prodrug, 5-aminolevulinic acid (5-ALA), which gets metabolized to PPIX intracellularly. Nevertheless, 5-ALA lacks tumor targeting properties, gets distributed among the whole body, and is excreted rapidly. To overcome such poor pharmacokinetics, conjugation to water soluble polymer-based drug delivery system was performed. Water soluble polymer-drug conjugates are known to be targeted to the tumor tissue by the enhanced permeability and retention (EPR) effect, which is based on the leaky tumor vasculature and absence of the lymphatic drainage within the tumor. An advantageous material for water soluble polymer conjugates is poly[N-(2-hydroxypropyl)methacrylamide] (PHPMA), mostly due to its biocompatibility and controlled synthesis.
