Cytoskeleton during the cell division cycle of the yeast Cryptococcus laurentii
Popis výsledku v původním jazyce
C. laurentii is occasionally diagnosed as an opportunistic infectious agent in immuno- suppressed patients, in the last years (1). The cytoskeleton of this yeast has not been studied yet. We examined the distribution and the dynamics of F-actin and microtubules in two strains of the yeast C. laurentii - encapsulated and non-encapsulated. The visualization of actin cytoskeleton by means of rhodamine-phalloidin in encapsulated strain C. laurentii CCY 17-3-5 failed. When observed, only the weak diffusionalfluorescence in the cells was identified. However, we detected the actin patches in the mother cells and their intensive fluorescence in the growing buds, which occured after application of monoclonal anti-actin antibody and consecutive secondary antibody SwAM-FITC. The second strain C. laurentii 17-3-6, which does not form capsule, was relatively more accessible for finding of F-actin using rhodamine-phalloidin. The cortical actin patches were detected.
