The production of hydroxyl radicals was evaluated as products of salicylic acid hydroxylation using HPLC with coulochemical detector. Two models were used. In human studies we evaluated the production of hydroxyl radicals in patients with cardiovasculardisease and in healthy volunteers. The determination of hydroxyl radicals in vivo was based on the evaluation of hydroxylation products of salicylic acid after the ingestion of acetylsalicylic acid (Aspirin). In animal studies we measured the productionof hydroxyl radicals in isolated rat and guinea pig hearts during ischemia-reperfusion injury. Isolated hearts were perfused with Krebs-Henseleit buffer containing 1mM salicylic acid according to the Langendorff method. The both methods above illustratea wide field of efficiency of HPLC methods coupled with coulometric detection for detection of the parameters of oxidative stress.
