The genomic landscape of uterine leiomyosarcomas has been increasingly defined in recent years with studies establishing inactivating alterations most often involving TP53, RB1 and/or ATRX [1]. In contrast, published genetic profiles of vulvar and vaginal leiomyosarcomas are limited owing to relative infrequency of these tumors. We therefore retrieved material from three spindled leiomyosarcomas that arose in the vulva or vagina, including one from our prior study [2], for next generation sequencing and MDM2 fluorescence in situ hybridization (FISH), the latter performed since MDM2 amplification has been reported in 3 of 80 uterine leiomyosarcomas or 4% of tested tumors [1]. We also evaluated a panel of immunohistochemical stains for expression of smooth muscle markers and hormone receptors.
