Název v původním jazyce
Study of genetic drift during conservation process of pea (Pisum sativum L.) genetic resources by microsatellite markers.
Popis výsledku v původním jazyce
In presented study, five accessions of pea (Pisum sativum L.) registered varieties (Bohatýr, Raman, Klatovský zelený, Arvika, Viktoria 75) were studied. Three temporally different samples composed of 20 individual plants for each accession, spanning theperiod of about 40-20-10 years and the most recent ones, were investigated and compared for 15 microsatellite loci. The aim of the study was: 1) to determine the effect of genetic drift during the process of maintenance and regeneration of accessions inex situ seed collections of genetic resources of pea, 2) study of intra-accession variation, mutability and speed of evolution of repetetive (SSR) sequences. In case of three accessions (Bohatýr, Arvika, Raman), evidence of genetic drift and intra-accession variation were clearly detected. In addition sequence analysis of 3 selected SSR loci in temporally different samples has revealed mutations of less stringently controlled repetetive motives.
