In the initiation phase of the project we reared and/or acquired approx. 3000 specific pathogen-free carp for experimental use. We used these fish to perform repeated intraperitoneal infection challenges with S. molnari blood stages and to monitor parasite proliferation over time using qPCR. Based on maximum parasite numbers in the blood, we have now established a continuous in vivo production line for parasite blood stages every 28 days (G2 – technically realized result/technicky realizované výsledky – prototyp, funkční vzorek) which allows us to be independent from the collection of environmental samples which is especially hindered in the winter months (frozen ponds, low temperature and undetectable parasite loads). Blood stages are harvested by centrifugation and used for in vivo and in vitro trials. We have performed a hematological study on carp blood and developed an immunoglobulin M sec qPCR assay to determine the immunological response of carp to parasite infection (in preparation for publication). Furthermore, together with Skretting ARC and Lallemond Animal Nutrition we have composed recipes for two carp diets, one based on curcumin (parasiticidal substance) and one based on yeast and probiotics (immunostimulants) to test these as an antiparasitic strategy in the newly established in vivo system.

Technické parametry
The presented functional sample represents a continuous in vivo culture and transmission system for blood stages of the myxozoan parasite S. molnari. S. molnari blood stages (SMBS) are transmitted to specific pathogen-free (SPF) fish via intraperitoneal injection and are harvested from host blood after 28 days, when parasite levels are at their peak. SMBS are then collected and used to infect a new lot of SPF carp. The aim of this process is 1. To be able to produce large numbers of SMBS throughout the year, independent from environmental conditions (frozen ponds during the winter months), 2. To work in a ‘clean’ culture system devoid of co-infections with other parasites, fungi or bacteria, usually present in ponds, and 3. To be able to test the effect of potentially parasiticidal or immunomodulatory substances in an in vivo model, aiming at essential reduction/removal of myxozoan proliferative stages. If effective, such substances could then be included into anti- parasitic diets for fish, with S. molnari serving as a model for other myxozoan infections in fish in aquaculture systems worldwide. This is of particular importance as a treatment against myxozoans in fish destined for human consumption is presently unavailable.
