Along with an expanding interest in proteomics in recent years, there has been a strong desire to develop methods permitting not only protein identification but also quantitation. Indeed, the quantitation of proteins in biological samples prepared upon different physiological states has become maybe the most important task in proteomics. Despite the progress, in mass spectrometry in particular, there is still no ideal method for protein quantitation because even the latest methods suffer from certain drawbacks. Currently, there are two main streams in quantitative proteomic: (i) the classical gel-based approach relies on protein staining in gel matrices, (ii) the second one, known as shotgun proteomics, employs mass spectrometry as a major tool for acquiring quantitative information. The latter utilizes mainly incorporation of stable isotopes into proteins or peptides, which are subsequently analyzed by mass spectrometry, however, one can take also advantage of label-free methods for s
