Detection of Resistance Genes in Solanaceae Using DNA Markers
Popis výsledku v původním jazyce
As model botanic species were selected tomato (Lycopersicon esculentum) and potato (Solanum tuberosum ssp. tuberosum). The potato was assessed for the collection of tetraploidic gene sources. The tomato was assessed for gene sources - varieties of the F1generation type and the segregating F2 generation. The potato's resistance against Ro1 of the Globodera rostochiensis pathotype was marked with a PCR amplification of the Gro 1.2.3 loci in chromosome II.. To assess the resistance, the results of experiments conducted by Leister et al. (1997) were used. One loci of quantitative resistant - PCR marker has a close gene link with the monitored resistance. For the dominantly established tomato's resistance against Meloidogyne incognita was used a co-dominant CAPS marker of the Mi gene. The experiments were based on tests by Williamson et al. (1994). Dominant and recessive alleles were detected after the digestion of the primary PCR product by the restriction enzyme TaqI. The identified PCR
