The evaluation of soil microflora activity was studied on defined localities with different nutrients contents and different yield potential at the experimental field (locality "Třebovle", Central Bohemia, Czech Republic). It was monitored during the years 2001-2002 as a part of the study for the development of precise farming systems there. The objective of this study was to evaluate: 1.effect of organic matter amendment on soil mineralization activity in vitro from two localities with different soil native P-content and pH; 2. microbial activity differences between spring and autumn soil sampling; 3. spatial variability within both sampling localities. Five different soil samples were taken from each of two localities (the first locality with lower soil native P-content and pH=5.4; the second one with higher soil native P-content and pH=6.9). Two variants (basal and potential) of respirometric tests (Filip 1997) were used for all soil samples. Carbon dioxide production by soil sample
