Molecular characterization of thermophilic and mesophilic Bacillus species

Molecular biology assays were used to determine the identification of thermophilic and mesophilic bacilli. A genotypic study using randomly amplified polymorphic DNA analysis (RAPD), amplification of the 16S-23S rRNA intergenic transcribed spacer (ITS-PCR), restriction fragment length polymorphism (RFLP) of 16S rRNA and restriction fragment length polymorphism of the ITS-PCR (ITS-PCR RFLP) was carried out with 5 thermophilic and 3 mesophilic Bacillus isolates. All above mentioned techniques have been optimized to be able to reach special electrophoresis pattern and discriminate among different species. RAPD protocol was optimized by using two different primers which both have been found to be effective for the RAPD fingerprinting of assessed bacilli. Amplification of the 16S-23S rRNA ITS produced multiple bands in gel electrophoresis, implying multiple copies of variable length in the ITS of these bacteria.
