Does oligo Tröger Base System Represent Novel Class of Heterocalixarenes? Synthesis and Conformation Study

At the end of 19th century, in 1887, Julius Tröger described that treatment of p toluidine with formaldehyde under acid condition forms a unique compound with nitrogens, which do not invert tetrahedral arrangement of moieties and pose as a chiral centers. The compound, today known as Tröger's base (TB), became as an example of "chiral nitrogen" stable at room temperature. At the end of 20th century supramolecular chemists brought up a certain renaissance of Tröger's base derivatives. The TB derivativesstopped being just a plaything of textbooks of the organic chemistry, and start to be a building block of molecular architects. The reasons for their rediscovery are the rigid V shape (85-120°), and of course, the inherent chirality. Thus TB derivativesfound usage in supramolecular chemistry, with advance for construction of receptor of nonchiral and chiral analyte, as well as in drug development.
