Characterization of temperature dependent release rate from liposomes

Liposomes are nanoscaled vesicles. They are able to encapsulate various types of substances because of phospoholipid bilayer that forms their shell. The membrane undergoes phase change at certain temperature that makes bilayer much more permeable than atlower temperatures. This phenomenon can be used for example in medicine for delivery of drugs. Liposomes can be imagined as small reservoir that can contain reactants that will be released from the vesicles after heating. Liposomes were prepared by hydration method using sonication, resp. extrusion as a homogenizing process. The vesicles have diameter of 50 nm to 200 nm depending on preparation conditions. 5(6)-carboxyfluorescein (CF) was chosen as a model substance. Amount of leaked CF was determinedby fluorescence spectroscopy. This work will show release rate studies of liposomes with different composition of their membranes meaning different temperatures of phase-change. The liposomes were made from dipalmitoylphosphatidylcholine
