UTILITY OF TARGETED NEXT-GENERATION SEQUENCING PANEL FOR COMPREHENSIVE FOCAL PROFILING OF DNA MUTATIONS IN TISSUE SAMPLES OF PATIENTS WITH IBD

  • Popis výsledku v původním jazyce

Development and progression of inflammatory bowel disease (IBD) is underlined by a number of molecular changes occurring within the affected tissue. The various types of reactive species arising during the inflammatory processes affect DNA within cells. The resulting DNA damage, mainly in form of somatic point mutations leads to various consequences, some of which may have profound effect on the course of the disease, therapy efficacy or, eventually, development of IBD-associated colorectal carcinoma. The molecular pathways of such malignant conversion exhibit distinct differences from mechanisms leading to sporadic types of colorectal carcinoma. The aim of the present work was to evaluate somatic DNA mutations occurring within the inflamed mucosa of patients with Crohn’s disease and Ulcerative colitis and compare those to mutation profiles obtained from sporadic cancers. We have targeted in a number of genes typically affected by somatic alterations in colorectal carcinoma pathways.
