Transplantation of stem cells represents a unique approach to treat spinal trauma. To follow in vivo traumatic changes, the signalling proteins were studied in the rat spinal cord transsection. 80th motoneurons and sensory neurons were subjected to analysis. Computer modeling of protein changes indicates that downregulated proteins belong to oxidative stress response as well as cellular fate and organisation. Increase of proteins involved in genome maintenance was observed in ventral horns. Only marginal changes were found in the dorsal root ganglia. To characterise mechanisms governing differentiation we combined the porcine neural stem cells in vitro differentiation and complementary proteomic analyses. The stem cells exhibited high levels of GRK2 and several members of FAS pathway. Typical for differentiated neural cells were enhanced integrin, PDGF and MAP kinase signalling and heme-oxygenase 2.
