The article deals with simultaneous use of the technique of relief contrast according to Hostounský (RCH – microscopy) and polarization microscopy to study the morphology of representatives of three classes of green algae (Cladophorophyceae, Zygnematophyceae and Charophyceae). Using this microscopic technique, new and interesting birefringent structures were found in all studied objects. This is mainly a finding of strongly birefringent cell walls of the filamentous algae of the genus Cladophora. In the algae of the genus Spirogyra, the cell walls were thinner and some of the younger individuals did not develop some transverse cell walls (no or very weak birefringence) which was evened out in later development. In the algae of the genus Chara, a completely new finding was the discovery of the presence of a significant amount of strongly birefringent particles in longitudinal rows in the surface layer of cells of the internode cortex. The technique simultaneous use of polarization and RCH-microscopy enable us to determine new and interesting birefringent structures in the examined biological objects.
