Systemic lupus erythematosus (SLE) is an autoimmune disorder which affects multiple organ systems and whose etiology still remains unknown. Nonetheless, many aspects of cellular and molecular interaction leading to the loss of self-tolerance have been clarified. The complement system plays a major yet ambivalent role in pathogenesis of systemic diseases especially SLE. On the one hand activation of the complement system leads to organ damage; on the other hand the complement system partakes in preserving the solubility of immune complexes and their transport to phagocytes of the reticulum-endothelial system and on the clearance of apoptotic cells from circulation. Both hereditary and acquired complement deficiencies are linked to various diseases. SLEcan be considered as a prototype of autoimmune disease, and can be used to demonstrate a variety of examples of complement dysfunction.
