Introduction: Sperm oxidative stress has been shown to detrimentally affect its lipid and protein content and to induce sperm DNA damage compromising sperm fertilizing ability. Superoxide, hydrogen peroxide and peroxynitrite are the common form of reactive oxygen species (ROS). Chemiluminescence using luminol measures both intracellular and extracellular ROS levels making it difficult to determine the source of the free radicals. Recently, flow-cytometry has been utilized to measure selectively intracellular ROS production with the added advantage of requiring smaller number of sperm. This study compared chemiluminescence and flow-cytometry in measuring sperm ROS production in a group of men within a large fertility clinic. Materials & methods: The study was approved by the Ethical Board of the Palacky University in Olomouc (CR). In order to included a broad spectrum of men the study recruited 166 men presenting with their partners for fertility assessment and 31 volunteers and sperm d
