The analysis of wheat genome could be simplified by its separation into individual chromosome and/or chromosome arms. The aim of this study was to develop an improved procedure for preparation of chromosome suspensions and to evaluate the potential of flow cytometry for chromosome sorting in wheat. Suspensions of intact chromosomes were prepared by mechanical homogenisation of synchronized root tip meristems after a mild formaldehyde fixation. The analysis of DAPI-staining chromosome suspensions resulted in flow karyotypes containing three composite peaks representing groups of chromosomes. In addition, a peak that represented chromosome 3B was well resolved. This chromosome could be sorted at high purity. Sorting of other chromosomes was not possible.The work is in progress to assess the suitability of various chromosome stocks to overcome this problem.
