Properties of an enzyme degrading cytokinins with isoprenoic side chain, previously named cytokinin oxidase were described and a new EC number has been preliminary assigned as 1.5.99.12. Cytokinin dehydrogenase is a 60kDa flavoprotein co-operating with some electron acceptor which features and localisation have not been elucidated yet. Enzyme has a crucial position in the metabolism of cytokinins, both endogenous or externally applied to plants. First structural cytokinin oxidase gene, ckx1, has been purified and characterised from maize in 1999. A family of seven genes has been found in Arabidopsis genome with high homology to the ckx1. We suggested several degenerated primers on the base of homology between Zea mays ckx gene and hypothetical genes from Arabidopsis. Poly+mRNA was isolated from barley and wheat grains and used as template for RT-PCR with all combination of primers.
