Morphological observation of bursa of Fabricius (BF) represent a valuable tool in evaluation of stress causative agents on poultry. The main goal of this study was to compare two methods for morphometry analysis of BF: standard hematoxylin/eosin (HE/EO) staining and immunofluorescence staining based on anti-basal cell cytokeratin antibody with DAPI background staining (CYT/DAPI). Chickens were divided into two groups (18 for HE/EO and 18 for CYT/DAPI). BF from 6 chickens from each group were taken in 7, 14 and 22 days of age. In every chicken, 40 randomly selected follicles were evaluated. In total, comparison of data from all days between groups didn't show significant differences. However, age have a significant impact. In 14 days of age, size of the follicle and cortex were significantly higher in CYT/DAPI group (P<0.05). Additionally, in 22 days old chickens size of the medulla was significantly higher in HE/EO group. Cortex/medulla ratio at age 7 days was significantly higher in HE/EO (P<0.05), but in older chickens were significantly higher (P<0.05) in CYT/DAPI group. Immunofluorescence staining seems to be more exact method and should be used, if precise localization of cortico-medullary border is necessary. On the other hand, HE/EO staining is a better choice for quantitative analyses of higher number of samples.
