Cats are the definitive hosts of Toxoplasma gondii. The distribution of genetically diverse T. gondii in cats is of importance to understand the transmission of this parasite. A 3-month-old domestic female cat was clinically examined because of lethargy, anorexia, fever, and diarrhoea. This cat was illegally transported from the Balkan area to the Czech Republic few days before occurrence of clinical symptoms. Numerous of Toxoplasma gondii/Hammondia hammondi-like oocysts were found in faeces; the cat was then medicated orally with toltrazuril (Baycox) and became asymptomatic after three days. T. gondii oocysts (designated Šimková isolate) were purified from faeces and genetic typed using the DNA extracted from tachyzoites isolated after bioassay in IFN-? knockout mice. 10 PCR-restriction fragment length polymorphism (RFLP) markers revealed Type III alleles at the SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, and PK1 loci and also Type III at the L358 and Apico loci. To our knowledge, this is the first T. gondii isolate genetically characterized from a clinically sick cat in the Czech Republic and the first viable European T. gondii genotype III in-vitro isolate from domestic cat.
