Generative reproduction of Potamogeton praelongus in laboratory is a possible way to preserve this critically endagered species in the Czech Republic and in the whole central Europe. Different ways of seeds storage, breaking seed's dormancy, cultivationtemperature, seeds' age and interactions of these factors were examined to determine suitable conditions for generative reproduction of P. praelongus in laboratory. The most suitable way of storage was keeping dried achenes in paper bags and submerging them in water for one month before beginning the test. Best results of breaking seed's dormancy were gained by near-natural methods - stratification, effect of microbial activity in water, scarification. Also methods using synthetic chemical substances (Savo Original, Ethephon, Gibberellic acid GA3) were successful. Multiple dormancy breaking measures proved successful. Seeds germinated better in a temperature-controlled incubator at 28 +- 1 oC in the dark than at room temperature 21 +- 3
