Given complexity and health impact of retinitis pigmentosa a better understanding of its pathogenesis is of major importance. Often used research models, such as mouse models, have been shown to insufficiently recapitulate the retinitis pigmentosa phenotype as it is observed in human patients. With the invention of induced pluripotent stem cells and the advances in retinal differentiation in the last decade new possibilities for retinal disease modeling have emerged. Two-dimensional and even three-dimensional in vitro models, known as retinal organoids, have been successfully derived from human stem cells. In this manuscript we describe and discuss the development of these stem cell derived retina models and their applications in retinitis pigmentosa research.
