Cancer causes the second largest number of deaths globally. Poor water-solubility of most anti-cancer agents and the involute fabrication process of many carrier-based drug delivery systems heavily hinder their clinical application. Here we report an easy manufactured nanoformulation based on both irinotecan hydrochloride and curcumin, which can be effectively self-delivered without utilising any carriers. This nanoformulation shows near-mono size distributions, tuneable surface charges and good stability under various conditions. Their effective anti-cancer efficacy, improved passive targeting and alleviated side effects are confirmed by in vivo tests on nude mice. This promising drug self-delivery system not only did solve the problems related to these two molecules, but also show it is efficient for tumour regression and multiple therapeutic functions to achieve ameliorated cancer therapy, with promise for clinical translation.
