    Gene expression control is fundamental to all life and can occur at several levels, with translation being one of the most crucial. Even a small disturbance in timing, spatial distribution and fidelity of translation of specific mRNAs causes or accompanies many diseases. Given that a) deregulation of expression of eIF3 subunits generates subcomplexes with altered repertoires of translated mRNAs affecting cell fitness, b) impaired eIF3 function is implicated in the etiology of various diseases including cancer, and c) observations that eIF3 can drive alternative modes of initiation in mRNA-specific manner, we consider eIF3 as one of the most intriguing subjects of study in this field. In this proposal, which is a continuation of our screening studies, we aim to investigate the eIF3 contribution to cell signaling and localized versus free protein synthesis by existing and newly proposed state-of-the-art techniques. A novel emerging pathway suggesting a cross-talk between DNA Damage Response and eIF3-driven translation with potentially exciting overlap to medicine will also be explored.
