Mitochondrial responses to seasonal cold acclimation and acute cold stress in Drosophila.

  • Annotation in Czech

This project will provide a comprehensive analysis of previously unknown adaptive changes in mitochondrial physiology that accompany the seasonal phenotypic switch in cold temperate and boreal insect species. Virtually everything we know about insect mitochondria relates to the active summer phenotype. Very little is known about how mitochondria switch from being an engine to support a vigorous summer metabolism (including flight) to a system to maintain a slow basal metabolism and to ensure survival at extremely low temperatures for long periods of the boreal winter. The project will compare changes in mitochondrial physiology induced by short photoperiod and cold acclimation in selected Drosophila species occurring beyond Arctic Circle with changes in tropical species and those that have recently colonised temperate zone. We will use an integrative approach combining microscopy of mitochondrial morphology, physiology of respiratory complexes, measurement of metabolic activities at tissue and whole organism levels, metabolomic and proteomic profiling, and functional experiments.
