Fluorescence microscopy is an essential tool for visualizing molecular processes in living systems. However, the full potential of fluorescent imaging remains unfulfilled due to intrinsic limitations of existing small organic fluorescent dyes (fluorophores), which often suffer from poor membrane permeability, non-specific interactions, and reliance on toxic, non-physiological synthetic methods. In this project, we propose to overcome these challenges by synthesizing state-of-the-art fluorophores directly inside cells through the bioorthogonal reaction of iminium salts (ImiSalts) with isonitriles. We will focus on optimizing the ImiSalt-isonitrile reaction, applying it in vitro for labeling of model proteins, and achieving real-time imaging in human cells. Success in this project will be an important contribution the field of molecular imaging. We anticipate that the proposed research will establish the ImiSalt-isonitrile reaction as a practical tool for studying biomolecules both in vitro and in vivo.
