Cílem projektu je analýza molekulární struktury lokusu 45S rDNA (ribozomální DNA) u banánovníku (Musa spp.). Poprvé bude určena úplná nukleotidová sekvence 45S rDNA u tohoto druhu a bude použita pro studium inzercí repetitivních sekvencí DNA do oblasti rDNA. Lokus rDNA u triploidního klonu s amplifikovanou oblastí organizátoru jadérka (NOR) bude použit jako model pro studium expanze rDNA lokusu. Znalost molekulární struktury oblastí IGS (intergenic spacer) umožní návrh molekulárních markerů, jejichž vhodnost pro fylogenetické analýzy bude ověřena na souboru vybraných druhů a klonů rodu Musa. Řešení projektu přispěje k objasnění evoluce této dynamické oblasti genomu a podstatně rozšíří dosud omezené poznatky o struktuře a evoluci genomu banánovníku. Poskytne rovněž cenné poznatky, které přispějí k sestavení úplné nukleotidové sekvence jeho jaderného genomu.

Project results evaluation
The structure of 45S rDNA was described in Musaceae. Phylogenetic reconstruction using ITS sequence showed that genus Musa is divided into two clades. Incomplete concerted evolution makes ITS analysis useful to determine genomic constitution in hybrids.
