Transport systems involved in intracellular pH homeostasis and yeast tolerance to low external pH
Annotation in Czech
We plan to participate in a COST Action, which aims to create a connected community of scientists working on the impacts of low pH (acid) on a range of important micro-organisms. In this project, we will use our expertise obtained and tools constructed so far to characterize in detail the relationship between various yeast transporters and yeast-cell tolerance to acidic environment. This new knowledge will contribute to the current state-of-the-art of molecular mechanisms underlying cation and pH homeostasis in yeast cells. Further, new knowledge will help to identify transporters whose engineering may improve the robustness of intracellular pH homeostasis together with the tolerance of yeast cells to growth-limiting external pH levels and/or elevated levels of organic acids in biotech-industry, or transporters as new targets for the development of antifungal drugs.

Project results evaluation
The project involved international collaboration and dealt with K+ and pH homeostasis in yeast cells upon acidic stress. A series of strains and plasmids suitable for studying changes in intracellular pH, membrane potential and cation homeostasis upon various, mainly acidic, stresses was prepared. New and highly-valued results were incorporated into five publications.
