Determination of pure muscle proteins by direct method

Project goals

The aim of the project is to select a direct laboratory method for determining the content of pure muscle protein (PMP) in meat products. In selected meat products, PMP analysis will be performed by a direct method of determining the creatinine content. The results of the analyzes will be verified by the indirect method based on the determination of the content of crude and pure proteins, as well as by the direct method of the determination of 3-methylhistidine and the NIR method. Changes in protein (proteome) content during the production of dry fermented meat products will be evaluated by advanced proteomic technologies based on mass spectrometry in order to reveal the effects of proteolysis during fermentation on PMP content.

    Možnosti stanovení čistých svalových bílkovin přímou metodou

    Cílem projektu je výběr přímé laboratorní metody pro stanovení obsahu čistých svalových bílkovin (ČSB) v masných výrobcích. Ve vybraných masných výrobcích bude provedena analýza ČSB přímou metodou stanovení obsahu kreatininu. Výsledky analýz budou ověřeny nepřímou metodou na základě stanovení obsahu hrubých i čistých bílkovin, dále přímou metodou stanovení 3-methylhistidinu a metodou NIR. Změny obsahu bílkovin (proteomu) během výroby fermentovaných trvanlivých masných výrobků budou hodnocené pokročilými proteomickými technologiemi na bázi hmotnostní spektrometrie s cílem odhalit dopady proteolýzy během fermentace a zrání na obsah ČSB.

    The project outputs were partially achieved, the V2-V4 (Nmet, Hneleg, Jost) results were achieved, while for the V1 (Jimp) result an extension of the deadline was approved. An extension of the deadline was justified and is relevant. Moreover, 11 additional results were produced beyond the plan (6x Jost, 2x Jimp, 3x O). The achieved results are of good quality, are related to the project objectives and will help disseminate the information on the determination of pure muscle proteins in meat products among the professional community. The results are of high quality and applicable in practice, by supervisory authorities in particular. As concerns the quality of project investigation, the results achieved and the fulfilment of the set out aim, this is an excellent project. The results achieved are of high quality and applicable in practice.
