Actinomycin D upregulates proapoptotic protein Puma and downregulates Bcl-2 mRNA in normal peripheral blood lymphocytes
Original language description
In PBL cultured in vitro the apoptosis induced by concentration of ActD causing almost exclusively damage of rDNA and subsequent ribosomal stress is dependent of p53 and Puma activation. The Bcl-2 mRNA downregulation is dependent of nucleolin cleavage byexecutive caspases. This apoptotic way, demonstrated in peripheral blood lymphocytes for the first time, is not the only way inducible in PBL by this cytostatic agent and by other genotoxic drugs
Czech name
Aktinomycin D indukuje proapoptotický protein Puma a reprimuje Bcl-2 mRNA v normálních periferních krevních lymfocytech
Czech description
V periferních krevních lymfocytech apoptoza indukovaná aktinomycinem D a následným ribozomálním stresem závisí na aktivaci p53 a Puma proteinů. Bcl-2 mRNA je redukována díky štěpení nukleolinu exekutivními kaspázami. Tato apoptotická cesta není jediná indukovatelná v lymfocytech aktinomycinem D či jinými genotoxickými látkami
