Limitation in reliability of fire blight (Erwinia amylovora) diagnostic methods
Original language description
A total of 20 Erwinia amylovora-like (Ea-like) isolates originating from symptomatic samples of fire blight host plants were analysed.Fourteen of them reacted negatively in immunochemical tests. Six Ea-like isolates reacted positively with a polyclonal antibody for PTA-ELISA (Loewe Co., Germany) and five of them gave positive reaction in indirect If test (Adgen co., Scotland). Three of those isolates were positive in a PCR test with primers AMSbL and AMSbR for E. amylovora. After modification of PCR protocol, putative strains reacted as negatives. Optimization of our PCR test was achieved by finding an optimal annealing temperature for primers. These three strains were identified using the microbial identification system BIOLOG as Pantoea dispersa andP.agglomerans. Identified strains were white variants of species P. dispersa and P. agglomerans that occur less frequently than yellow variants.

Omezení v spolehlivosti diagnostických metod pro Erwinia amylovora
