    Thermal analysis for cryopreservation method improvement

    Changes in thermal characteristics of plant tissues are important for explants successful cryopreservation. These changes can be influenced by genotype of plant, preculture conditions of explants and osmotically active solution used. The effect of genotype was studied in three grapevine varieties with different sensitivity to dehydration. Different sensitivity of explants to dehydration influenced significantly their tolerance to cryopreservation. A significant effect of explants acclimation on their tolerance to dehydration before cryopreservation was detected in poplar explants. Low temperature treatment resulted in a significant increase in poplar explants tolerance to dehydration. In addition, the positive simultaneous effect of low temperature and osmotic treatment by 0.7M sucrose on explant tolerance to dehydration was demonstrated. Thermal analysis allowed control of the whole dehydration process, including optimal hydration level of explants before cryopreservation.
