Assessment of retinoic activity in fly ash and air samples from polluted areas using P19/A15 cell line
Original language description
Contamination in the environment has been described to induce malformations in amphibians. This fact could be linked with anti/retinoic action of the contaminants because retinoids control morphogenesis by affecting processes such as differentiation or apoptosis. In vitro model for evaluation of anti/retinoic action of xenobiotics was developed. It is based on murine cell line P19, which retains a pluripotent character and its differentiation is regulated mainly by retinoids. In the derived cell line P19/A15, the retinoic response element was linked with luciferase gene enabling the study of retinoid signaling. We tested air samples extracts from two areas in the vicinity of chemical factories producing different types of pollutants, extracts from flyash from municipal incinerator and reference sample from background locality. Important activities were observed in both gas and particulate fractions of multiple air samples and in fly ash samples.
