Gradient elution improve peak capacity with respect to isocratic conditions and should be preferably used in 2D LC x LC in both dimensions. Gradient conditions spanning over a broad range are used with generic "full in fraction" (FIF) gradients. "Segmentin fraction" (SIF) gradients cover a limited gradient range. Optimization of the gradient profiles is important tool for achieving high 2D peak capacity and savings of the separation time. The transferred fraction volumes may significantly affect the bandwidths, especially at high elution strength of the fraction solvent, which may cause even band distortion or splitting in combined normal-phase (HILIC)-RP systems, but also in some 2D RP-RP systems.
