The Czech Crop Cryobank stored in total more than 500 accessions of 14 different crop species. The aim of this work was to determine the effect of dehydration on the pollen and explant thermal properties and their vitality in potatoes and hops. Explant survival and regeneration ability after cryopreservation was affected by genotype, significantly increased by osmotic stress, and depended on dehydration using the modified cryoprotective solution PVS3. The optimal explant dehydration time (60 minutes) led to a significant decrease in free water content to 0.3–5 %, while further dehydration led to a decrease in their vitality. When pollen water content failed below 0.27 g of water per g of dry matter, water does not crystallize and at the same time, the presence of a glass transition has always been demonstrated. With strong dehydration of pollen to 0.1 g of water per g of dry matter, the glass transition temperature increased to -5 °C, which represents increase in the safe storage temperature of pollen as well. Results showed that a suitable method of dehydration can limit the proportion of frozen water in explants in tested species of plants and the observed thermal properties of pollen confirm the suitability of the cryopreservation method for their long-term storage in liquid nitrogen conditions.
