Acetylcholinesterase (AChE) is an enzyme playing a pivotal role in cholinergic neurotransmission. It is inhibited by a wide group of neurotoxic compounds. Owing to the practical significance, pesticides (e.g. carbaryl, dichlorvos, dimethoate) nerve agents (e.g. sarin, soman, tabun, VX) natural toxins (e.g. aflatoxins from Aspergillus flavus) and drugs (e.g. Alzheimer´s disease drugs galantamine, rivastigmine, donepezil) can be example as the AChE inhibitors. The neurotoxins can be easily assayed by devices containing AChE as a recognition element. Several colorimetric dipsticks and biosensors are typical tools containing AChE. The reported experiment is devoted to the construction of dipstick with immobilized AChE and indoxylacetate respective 2,5-dichlorophenol acetate as substrates. Original idea is to construct devices providing a contrast coloration comparing to the commercially available devices based on color change white to yellow as they use Ellman´s reagent.
