Analytical performances of validated chemiluminescent enzyme immunoassays to detect N-methylcarbamate pesticides.
Original language description
In the present work, the comparison study of enzyme-linked immunosorbent assays (ELISAs) with colorimetric detection and ELISAs with chemiluminescent detection for the determination of carbofuran, carbaryl and methiocarb is described. Both conjugate-coated formats are based on identical monoclonal antibodies and homologous protein conjugates. The sensitivity of immunochemical method was expressed as a detection limit, a linear working range and an I50 value. In comparison with colorimetric ELISA, the ability of the chemiluminescent technique to detect lower concentration of HRP allowed to decreased the optimal antibody and conjugate concentrations and thus to reach lower analytical parameters. The experimental comparison of the analytical performance of ELISAs was carried out by simultaneous analysis of apple-strawberry baby food extracts spiked with mentioned pesticides at three different concentration levels. The extracts were prepared in two duplicate sets - crude extracts and extra
