Bacteria of the Acinetobacter genus belong to the Gram-negative, non-spore-forming, non-motile, non-fermentative, oxidase-negative, nitrate-negative, strictly aerobic, bacteria. There are important soil and water bacteria with wide occurrence in nature, raw food materials, foods and human sources. Acinetobacteria represent health risk bacteria causing in immunocompromised individuals life-threatening infections, mostly nosocomial infections. These bacteria represent a significant technological risk in the food industry where causing serious problems in connection to many sensory and texture defects. They can be detected relatively quickly with high reliability at the genus and species levels using different methods. The aim of this work was the identification of Acinetobacter spp. strains, which were isolated from raw material, food products and production facilities, using the PCR method with newly designed genus-specific primers. First, isolates were characterized by various microbiological and biochemical tests. Their pre-identification was done by use of the MALDI-TOF MS method and reliable identification was done using of the 16S rRNA sequencing analysis; by both methods at the species level. It was found that the isolates belonged to the six different Acinetobacter spp. (A. baumannii, A. calcoaceticus, A. guillouiae, A. johnsonii, A. lwoffii and A. pittii). Further it was found that individually designed genus-specific AcinetoFor1 and AcinetoRev1 primers were 100% specific for genotypic identification of bacteria of the Acinetobacter genus using the PCR method. Obtained results may have been useful for assurance safety and quality of food raw materials and final food products using reliable and rapid identification method.
